I've used 5.0 grains of Bullseye, 7.5-8.0 grains of Unique, 6.5 grains of W231, and 5.5 grains of Titegroup with either a handcast Lyman 454190 (251 grain RNFP) or a handcast RCBS 45-255KT (262 grain SWC) with good results. All of these chrono between 700 and 825 fps. The very first time I used Titegroup, I had a squib toward the end of the first 50 round box where the powder charge burned and the round never made a sound, I thought it was a dead primer. Really weird, lotsa burnt residue in the case when I pulled the bullet it didn't generate enough pressure to even unseat the bullet.
Now for Ruger ONLY, I load 24 grains of H110 or W296 under the 262 grain cast SWC for ~1350-1400 over the chrono, don't use this in a Colt or clone or you'll turn it into a 3 shooter whose cylinder won't turn after the first shot.

I've had very good results with a 250g Laser Cast bullet over 9.0g of Unique for my 45 Colt Anaconda. I've had good groups using 6.2g of Titegroup for my SAA.
I had great goups with a 260g Speer HP over 6.5 grains of HP-38 for the Anaconda, though I'm sure it wasn't going over 750 fps.

I like that Anaconda! Bought a .44 version last year for $399.00 and then Colt made the announcement that they were dropping all their DA line and a couple months later I saw a .45 version advertized in the local rag for a whopping $980.00!! I'd like to find a .45, but not at that price!
